Labbadia takes over from Michael Wimmer who had acted as caretaker after Pellegrino Matarazzo had to go in October.
He will begin training with the team on December 12, the club said on Monday.
"Stuttgart is not just any club for me. I spent a very important phase of my coaching career in Stuttgart over almost three years and now I want to help the team stay in the Bundesliga," Labbadia said.
Former Germany striker Labbadia's first term in Stuttgart was between 2010 and 2013. He last coached at Hertha Berlin for nine months in 2021.
The veteran has also coached Hamburg SV, twice, Bayer Leverkusen, Wolfsburg, Darmstadt and Gruether Furth.
The club had on Saturday also filled the vacant sporting director position with Fabian Wohlgemuth, after parting ways with Sven Mislintat earlier last week.
"We need experience and stability now. I'm therefore delighted that we have signed Bruno Labbadia as head coach," Wohlgemuth said.
The Bundesliga resumes after the World Cup and winter break in mid-January, with Stuttgart starting at home against Mainz on January 21.
