Detectives say a member of the public saw what they believed to be a body in the river near Morpeth just after 5.30pm.
Officers from Central Hunter Local Area Command attended the location and established a crime scene.
An investigation was commenced and the body retrieved for examination by specialist forensic officers.
The deceased man has not been formally identified, but police say their preliminary findings indicate the man's death may not be suspicious.
Anyone with information that may be able to help investigators establish how the man ended up in the Hunter River is asked to contact CrimeStoppers.
A report is being prepared for the Coroner.
