The New England Highway is closed in both directions at 3:30pm on Sunday and motorists are urged to delay their travel plans and avoid the area.
At 3:25pm Westpac helicopter rescue spokesperson, Glen Ramplin, spoke to the Hunter Valley News from the accident scene.
He said the single occupant of a Citroen, a man believed to be in his 60s, had just been released from the wreckage of one vehicle with multiple injuries.
Mr Ramplin said Westpac’s Rescue Three helicopter, based in Tamworth, is airlifting the patient to the John Hunter Hospital in Newcastle.
The two occupants of a Toyota Prado, a male and a female, were still being attended to by rescue workers.
Mr Ramplin said Westpac’s Rescue One helicopter, based in Newcastle, was on standby at the scene to transport the injured couple to the John Hunter Hospital.
It is understood their injuries are not life-threatening.
Police, highway patrol, paramedic and fire emergency services are in attendance.
The accident occurred near the township of Wingen in a 100km/h speed zone area around 1:50pm on Sunday.
The Transport Management Centre said it is using its digital Variable Messaging Sign system to alert motorists of the New England Highway closure at Wingen.
A spokeswoman said a sign is operating south of Maitland at Black Hill on the M1.
Another sign is also warning of the New England Highway closure south of Port Macquarie at Lake Innes on the Pacific Highway.
Motorists are urged to heed these warnings until further information can be obtained from police, rescue crews and traffic control management at the scene.
